Create, detach, and reattach a forward primer

Create and save a forward primer from a selection

Start with a nucleotide sequence in a writable local project:

The Primers drawer opens beside the far-right sequence tool rail. Its toolbar entry is the fourth item under Tools, below Annotations.

1. Open the nucleotide sequence and select 2D.
2. Drag across the visible sequence canvas to select the intended binding region.
3. Right-click the selection.
4. Select Create Forward Primer →.
5. When the primer form opens, enter a name.
6. Review the inline save location. By default, DNAForge uses the folder that contains the open sequence.
7. Select Save Primer.

DNAForge closes the form and saves a .primer.json record with one binding to the source sequence. Attached becomes active and shows the named primer. Saving the primer does not change the source sequence file.

To discard the draft before saving, select Cancel. DNAForge closes the form without writing a primer record or changing the source sequence.

Detach the saved primer

1. Select Attached at the top of the Primers drawer.
2. Find the saved primer.
3. Point to its row to reveal the available actions.
4. Select Detach.

DNAForge removes the binding to the open sequence. The saved primer remains available to the Find workflow below.

Find and reattach the saved primer

1. Select Find at the top of the open Primers drawer.
2. Set the search region to Entire sequence.
3. Select Find Primers.
4. Wait until the saved primer's Attach control appears.
5. Select Attach.
6. Select Attached and confirm that the saved primer is listed again.

DNAForge adds one binding between the saved primer and the open sequence. It does not add the primer as an annotation inside the source sequence file.
